One of the characters in Takemiya Jin's Fragments of Love has a brother, and part of the story is him discovering his sister's homosexuality as well as the reciprocal. I'm not sure he's a younger brother, though.

I mean that usually there aren't siblings. If they are - it's usually a younger/older sister - can be also a twin. And in many cases it's a part of a plot for incest yuri.

Probably not what you're looking for, but Mariaana in Transistor Venus has a younger brother. There's a chapter where Enus is visiting them, but I forgot which one it is. It's the one where we learn about Enus intriguing theory that everybody has a limited number of kisses during their life and therefore they shouldn't be used up by kissing siblings (in a non-incestuous way ^^).
