I don't think the sibling is trans, they look like a butch / masc lesbian to me and the dialogue from the father seems realted to becoming a butch / masc lesbian

It doesn't make sense for the sibling to be trans just to be with woman but does make sense that they are a lesbian and are becoming butch / masc to be with their girlfriend

Transgender people transition because of gender dysphoria
