Basically all 4 (human) members of 25-ji are being psychologically tortured for something they are unable to change, cannot be who they want to be, cannot do what they want to do, and are in the state of "to live is pain" and "desire of disappearing from the world". Mafuyu being somewhat a more serious one, having no emotion or desire for herself and prob. has a death wish or so (and frequently depicted in fanart as such).
