Yes it's canon, Velma is outright stated to have a crush on Coco Diablo, the designer of all the monster suits bad guys use to commit crime, and Coco flirts back. And Daphne plays wingman, it's cute.

Its kind of weird to think about how Velma being a lesbian was essentially memed into cannon. It wasn't like there was subtext in the original Scooby-Doo cartoon or anything, people were just like "lol Velma looks like a lez" as a joke as far as
I was aware.
Its like if they canonically made He-Man homosexual because of all the jokes people made about that cartoon.

Velma was at one point played by Hayley Kiyoko in one/ some of the live action movies so it was inevitable that Velma'll be gay.

The Mystery Inc. version of Velma is also canonically lesbian, although she only ever dated Shaggy in that show

^Wouldn't that make her Bi? I never did watch the show in its entirety but I do remember her being super into Shaggy in what I saw.
